Moving from three litter boxes to two?

I have two cats and the recommended three litter boxes, two downstairs in different rooms and one upstairs. The upstairs one was originally used by the youngest cat when he was shut in that room during the introductions phase between my two cats. They both now roam freely and the upstairs box hasn't been used in at least six weeks, even though my eldest cat spends lots of time upstairs she always comes downstairs to use one of those boxes, and the younger one rarely goes upstairs.

Can I get rid of the upstairs box?

I have three cats with two boxes. They just never used the third.

If it hasn't been used in weeks, I would go ahead and remove it.

If they haven't used the upstairs box in weeks, I think you can remove it. I had the "requisite" one box per floor for our last cat and ended up removing one because he never used it. When he was a senior with heart disease and going up and down the stairs had become a bit of a chore for him, I put it back and he used it.
